DualityBio (映恩生物)
Shanghai ADC developer founded in 2019 that out-licensed most of its pipeline to global pharma and listed in Hong Kong in 2025.
1. Core Product / Service
DualityBio (Duality Biotherapeutics, HKEX: 9606) develops antibody-drug conjugates (ADCs). It runs four in-house ADC platforms, called DITAC, DIBAC, DIMAC and DUPAC. As of early 2026 it had 10 clinical-stage assets and more than 3,200 patients enrolled worldwide, about half of them outside mainland China [3][4].
- DB-1303 / BNT323 (HER2 ADC): China BLA under review for HER2+ breast cancer [3][4].
- DB-1311 / BNT324 (B7-H3 ADC): entering a global Phase 3 trial in prostate cancer. Its B7-H3 antibody is in-licensed from WuXi Biologics [2][3].
- DB-2304 (BDCA2 ADC): Phase 2a in lupus, an autoimmune use of ADCs [3].
- Bispecific ADCs: DB-1418 (EGFR×HER3, partnered with Avenzo) and DB-1419 (B7-H3×PD-L1) [3].
2. Target Users & Pain Points
Its main customers so far are licensing partners, not patients. In 2025, 99.86% of its RMB 1.85B revenue came from licensing and collaborative R&D services [5]. Partners get China-generated clinical data and global trial execution for ADCs at a speed and cost that would be hard to match in-house.
3. Competitive Landscape
| Player | Positioning | Vs. DualityBio |
|---|---|---|
| medilink-therapeutics | Suzhou ADC developer, founded 2020; large Roche and BioNTech deals | Similar BD-funded model; got a single larger upfront (YL201) |
| minghui-pharmaceutical | Shanghai/Hangzhou ADC and bispecific developer | Smaller BD (China-only Qilu deal); still pre-IPO |
| Large Chinese ADC houses | Integrated developers with their own plants | DualityBio stays asset-light on manufacturing |
| allink-biotherapeutics | ADC startup founded in 2023 | Same modality, about four years behind |
4. Unique Observations
- Wet-lab model: the prospectus is explicit. DualityBio keeps R&D in-house, with 131 of its 170 employees in R&D at end-2024. It works with more than 200 CROs and outsources all manufacturing to six CDMOs in China. Only 4 employees were in manufacturing and quality control. It leases about 5,039 m² across Shanghai, Jiangsu and Beijing, and US staff work remotely [2]. Its 2025 Lingang drug manufacturing licence is local production through partners, not an owned plant [5].
- BD funds the company: it has reported positive operating cash flow for three straight years, which is rare for a pre-revenue biotech [3].
- Founder is an investor: Zhu Zhongyuan came from venture capital (6 Dimensions, WuXi Healthcare Ventures), not from a pharma R&D career [2].
