Junevity

San Francisco UCSF spinout that uses ML to pick transcription-factor targets and drugs them with twice-yearly siRNA; lead in T2D/obesity.

1. Core Product / Service

Junevity's RESET platform combines human disease omics, genetic associations and machine learning. Its purpose is to find transcription factors whose activity drives a cell's damaged state; the company then silences them with siRNA to "reset" cells to health [1][3]. The underlying research came from co-founder Janine Sengstack's work at UCSF and is exclusively licensed [1].

The lead candidate JUN_01 targets a confidential transcription factor linked to metabolic dysfunction, insulin signaling and appetite. It is being developed for type 2 diabetes and obesity, dosed once every six months [3]. The company reports preclinical improvements in blood glucose and insulin sensitivity, weight loss with preserved lean mass, and resistance to weight regain; these are company data [3]. JUN_01 is in IND-enabling work, with first-in-human studies planned for the second half of 2026 [3]. Early neurodegeneration programs are planned [3].

Wet lab. Undisclosed. No lab location, area, job postings or CRO partners were found. A database lists a San Francisco address and roughly 13 employees; both are unverified. AI is used for target selection, and siRNA is a mature modality with an established outsourced chemistry and toxicology supply chain. It is therefore plausible that most synthesis and IND-enabling studies are outsourced, but this is an inference, not a disclosure.

2. Target Users & Pain Points

  • Patients: people with type 2 diabetes and obesity who want durable metabolic control and weight maintenance without weekly injections.
  • Pain addressed: incretin drugs need frequent dosing and weight often returns after stopping. A twice-yearly siRNA acting on a regulator upstream of many pathways is the company's alternative [3].

3. Competitive Landscape

Player Approach Vs. Junevity
kailera-therapeutics / verdiva-bio Next-generation incretins Established mechanism; frequent dosing
Alnylam, Arrowhead siRNA metabolic programs Mature siRNA platforms with known targets
newlimit Transcription-factor reprogramming via mRNA-LNP Same "cell reset" idea, delivered as gene expression rather than silencing
retro-biosciences / altos-labs Rejuvenation biology Much larger capital; longevity framing

4. Unique Observations

  • Moderate AI weighting. AI selects targets; the drug format, siRNA, is well proven. Junevity is therefore less "AI-native" than de novo design companies, but its clinical risk is concentrated in target biology rather than in an unproven modality.
  • Fast to clinic for its size. With a $20M seed, the company plans first-in-human about three years after founding. That is faster than most US AI biotechs founded in 2023 or later, apart from the much better-funded Mirador.
  • Longevity framing, metabolic first indication. The company launched as a "cell reset for longevity" story, but its first indication is a conventional, large metabolic market [1][3].

5. Financials / Funding

Round Date Amount Investors
Seed 2025-02-13 $10M Goldcrest Capital, Godfrey Capital (leads) [1]
Seed expansion 2025-12 $10M (seed total $20M) Goldcrest, Godfrey [2][3]
  • Total: $20M [2]. The CEO said the company was not actively fundraising when the expansion came in [2].

6. People & Relationships

  • Co-founder & CEO: John Hoekman (co-founder of Impel Pharmaceuticals) [3].
  • Co-founder & CSO: Janine Sengstack (UCSF research basis) [1].
  • COO: Rob Cahill [1]. One profile also lists Hao Li as a founder (unverified).
  • Origin: UCSF spinout, founded 2023 [3][4].
  • Peers: newlimit, retro-biosciences, kailera-therapeutics.
