Monte Rosa Therapeutics
Boston/Basel molecular-glue degrader company; AI/ML-enabled QuEEN engine, VAV1 glue in Phase 2 with Novartis.
1. Core Product / Service
Monte Rosa's Swiss operating company was incorporated in Basel in April 2018, and its Delaware parent in November 2019. Headquarters are in Boston [1]. It discovers molecular glue degraders (MGDs), small molecules that make the cereblon E3 ligase tag a target protein for destruction. Its QuEEN engine combines "AI/ML engines with multiple proprietary experimental tools". These include a geometric deep-learning model of protein surfaces that mines sequence and structure databases for degron-like features, and AI models that "learn from and guide our high throughput screening and chemo-proteomics" [1].
Pipeline (Oct 2026) [1][2][4]:
- MRT-6160 (VAV1 MGD): licensed to Novartis, which started a Phase 2 in Sjögren's disease and paid a $50M milestone (Sept 2026).
- MRT-8102 (NEK7 MGD, NLRP3/IL-1 pathway, wholly owned): positive Phase 1 in obese subjects at high cardiovascular risk (Oct 2026; median hsCRP −85%). Phase 2 trials in coronary disease, gout and hidradenitis suppurativa are planned for late 2026 to 1H 2027.
- MRT-2359 (GSPT1 MGD): Phase 2 with apalutamide in prostate cancer, activated Aug 2026.
Wet lab. Monte Rosa is lab-heavy. It leases ~63,300 sf of office and lab space in Boston and ~44,700 sf in Basel (lease extended to June 2028), about 108,000 sf in total [1][2]. The 10-K describes in-house high-throughput assays "coupled with customized automation and robotic systems" for ternary-complex and degradation readouts, plus quantitative mass-spec proteomics, chemoproteomics and structural biology. The MGD library holds 75,000+ compounds [1]. Headcount was 150 at end-2025 (118 in R&D), up from 134 [1]. Lab equipment at cost was $27.0M [1]. Drug substance and product are made by third-party CMOs on purchase orders, and CROs run studies. The company also discloses "a supplier based in China" that supports MGD library expansion and MRT-2359, without naming it [1].
2. Target Users & Pain Points
- Patients: immunology (Sjögren's, inflammatory diseases), cardiometabolic inflammation and oncology.
- Partners: Novartis (VAV1 licence; separate I&I deal) and Roche (oncology/neurology MGDs) use QuEEN to reach targets that lack ligandable pockets.
- Pain addressed: rational molecular-glue discovery was historically serendipitous. QuEEN aims to make degron matching systematic.
3. Competitive Landscape
| Player | Approach | Vs. Monte Rosa |
|---|---|---|
| schrodinger | Physics + ML small-molecule design | Software-led; does not focus on degraders |
| recursion-pharmaceuticals | Phenomics + automated chemistry | Broader platform, larger lab footprint |
| iambic-therapeutics | AI-central small molecules | AI is the core claim; Monte Rosa leads with chemistry and proteomics |
4. Unique Observations
- AI as a layer on a wet-lab engine. The disclosed moat is the MGD library, proteomics and automated screening. The filings position AI as connecting those assets, not replacing them [1].
- Partnering funded the build. About $370M of non-dilutive upfronts and milestones from Roche and Novartis sits alongside ~$0.95B of equity. That puts Monte Rosa among the better-capitalised companies founded in 2018–2022 (cash $621M, runway into 2029) [1][2].
- "US-based" is partly a domicile question. The science started in Basel, and the Basel lab remains roughly 40% of the footprint.
5. Financials / Funding
- Series A $32.5M, B $96M, C $95M (2020–21); IPO June 2021 $255.6M gross at $19 [5].
- Follow-ons: $24.9M (2023), $96.4M (2024), and $345.0M gross in Jan 2026 at $24 [1][3]. Cumulative equity is about $0.95B excluding ATM sales.
- Non-dilutive: Roche $50M upfront; Novartis $150M + $120M upfronts and a $50M milestone [1][2].
6. People & Relationships
- Founders: Versant Ventures with Raj Chopra and Ian Collins (Institute of Cancer Research, UK) [5].
- Management: Markus Warmuth (CEO; ex-H3 Biomedicine), Sharon Townson (CSO; ex-Kymera), Filip Janku (CMO; ex-MD Anderson) [1].
- Partners: Novartis, Roche.
