Eikon Therapeutics

Imaging-first oncology biotech: live-cell single-molecule tracking plus ML, ~$1.5B raised, lead in-licensed TLR7/8 drug in Phase 2/3.

1. Core Product / Service

Eikon was incorporated in July 2019. Its discovery engine is a proprietary single-molecule tracking (SMT) platform: custom super-resolution microscopes image how individual proteins move inside living cells, robotic high-throughput screening runs around the clock, and machine-learning models analyse the trajectories (diffusion, dwell time, clustering) to find compounds that change protein behaviour [1]. The company claims each microscope captures over 100,000 protein-motion events in 1.5 seconds and the fleet produces up to ~1 PB of data per day [1].

The pipeline is oncology-focused and mixes in-licensed and internal assets [1][2]:

  • EIK1001 (IV TLR7/8 co-agonist; in-licensed in 2023 from Seven and Eight / Birdie, China): two registrational Phase 2/3 trials, in 1L melanoma with pembrolizumab and in 1L NSCLC. After the first interim analysis in Aug 2026 the data monitoring committee recommended continuing the melanoma trial.
  • EIK1003 / EIK1004 (PARP1-selective; in-licensed from Impact Therapeutics, Shanghai): Phase 1/2.
  • EIK1005 (WRN helicase, internally discovered): entered Phase 1/2 in Jan 2026. EIK1006 (AR antagonist, internally discovered): IND guided for end-2026 to Q1 2027.

Wet lab. Eikon runs one of the largest in-house discovery footprints among AI-labelled biotechs. It occupies a 285,000 sf headquarters in Millbrae, CA (moved in April 2025, lease to 2040) and holds ~13,000 sf of R&D space in New York [1]. About 125,000 sf in Hayward was vacated in 2025 and impaired [1][2]. Headcount was 384 at end-2025, 321 of them in R&D [1]. Net PP&E was ~$143M, mostly leasehold improvements ($121M) plus ~$26M of lab equipment [1]. Manufacturing is fully outsourced: the company does not own or plan any manufacturing facilities. Together with licensor Impact it uses WuXi AppTec for selected in vivo pharmacology, CMC and nonclinical testing, and WuXi is involved in clinical supply for its clinical programmes [1]. Global trials run through CROs that are not named.

2. Target Users & Pain Points

  • Patients / own pipeline: melanoma, NSCLC, PARP1- and WRN-dependent tumours, prostate cancer.
  • Pain addressed: static structures and endpoint assays miss how drugs change protein dynamics in live cells. SMT is meant to read that directly at screening scale.
  • Eikon has no platform-partnership revenue. An external research-access programme for its instruments was the unit cut in the May 2025 layoff [3].

3. Competitive Landscape

Player Approach Vs. Eikon
recursion-pharmaceuticals Phenomic imaging HTS + ML Closest in lab intensity; Recursion images cell morphology, Eikon images single-molecule motion
insitro iPSC + pooled CRISPR + ML Biology-first data engine, earlier pipeline
iambic-therapeutics AI small-molecule DMT loops ~12.5k sf lab today vs Eikon's ~285k sf
generate-biomedicines Generative biologics, ~146k sf Biologics; similar "own the data factory" stance

4. Unique Observations

  • The platform and the late-stage pipeline are decoupled. The three most advanced assets were all in-licensed from Chinese biotechs. Only EIK1005 and EIK1006 are platform-derived, and both are early. The value of the IPO therefore rests mainly on EIK1001, not on SMT [1].
  • Money is moving from platform to clinic. FY2025 R&D was ~$250M, split ~$130M research/engineering and ~$117M clinical. In 1H 2026 research/engineering fell 16% while clinical spend rose 53% [1][2].
  • Heavy fixed footprint at a small-company headcount. About 285k sf for 384 people is roughly 740 sf per employee, above the 450–600 sf range seen at peers such as generate-biomedicines and iambic-therapeutics's new site.
  • AI is a layer, not the thesis. The 10-K calls Eikon "a late-stage clinical biopharmaceutical company". ML analyses imaging data and is not marketed as the core asset [1].

5. Financials / Funding

  • Series A $148M (2021); Series B $517.8M (Jan 2022); Series C ~$106M cash (2023); Series D $350.7M (Feb 2025, existing investors, structured with preferred exchanges and warrants) [1][4][5].
  • IPO on Nasdaq as EIKN, Feb 2026: $381.2M gross at $18 per share, upsized [6]. Cumulative raise is roughly $1.5B.
  • Cash $531.2M at 2026-06-30, guided into 2H 2027. Q2 2026 R&D spend was $75.5M [2].
  • May 2025 reduction in force: 55 roles (~15%) [3].

6. People & Relationships

  • Founders: Eric Betzig (2014 Nobel, super-resolution microscopy), Robert Tjian, Xavier Darzacq, Luke Lavis. The 10-K credits only Tjian and Betzig [1][5].
  • Management: Roger Perlmutter (Chair/CEO; ex-Merck Research Labs, ex-Amgen R&D), Roy Baynes (CMO), Russ Berman (CTO) [1].
  • Licensors: Seven and Eight / Birdie (EIK1001), Impact Therapeutics (EIK1003/1004).
  • Peers: recursion-pharmaceuticals, insitro, schrodinger; 2018–2022 cohort peers treeline-biosciences, odyssey-therapeutics.
