Genesis Molecular AI

Bay Area AI small-molecule company (formerly Genesis Therapeutics); Pearl structure model, Incyte/Gilead deals, preclinical PI3Kα lead.

1. Core Product / Service

Genesis was founded in 2019 as a spinout of Stanford's Pande lab. It renamed itself from Genesis Therapeutics to Genesis Molecular AI on 28 Oct 2025 [4]. Its GEMS platform is pitched as an "AI operating system for molecular design" that combines foundation models with physics [1]. In Oct 2025 it released Pearl, a protein–ligand co-folding model trained heavily on physics-generated synthetic data, which the company claims outperforms alphafold3 on its benchmarks [4]. Later releases include a distilled faster variant and an internal agentic discovery system (single-source).

The internal pipeline is entirely preclinical [1]:

  • Lead: a pan-mutant PI3Kα allosteric inhibitor for breast and colorectal cancer, "approaching development candidate nomination".
  • A cell-death (extrinsic apoptosis) programme.
  • Several immunology small-molecule programmes.

Most revenue so far comes from partnerships. Upfront payments total $185M: Incyte $150M across two deals and Gilead $35M [1][2][3].

Wet lab. Genesis runs a "fully integrated laboratory" in San Diego; AI teams sit in the Bay Area and New York [1]. Job posts confirm in-house medicinal chemistry, computational chemistry, biology, DMPK and a CMC director role [9]. Lab size and address are not disclosed, and headcount is roughly 145–160 by third-party counts [8]. Chemistry postings refer to "industry collaborators" but name no CRO [9]. Under the 2026 Incyte expansion, Incyte shares proprietary experimental data with Genesis [2], so partner wet-lab output also feeds the models.

2. Target Users & Pain Points

  • Pharma partners: Incyte (at least seven targets across two deals) and Gilead (three targets, with options). Earlier deals with Genentech and Eli Lilly (2022: $20M upfront, up to $670M) are no longer listed as active [1][3][8].
  • Own pipeline: oncology and immunology targets that are hard to drug with standard medicinal chemistry.
  • Pain addressed: structure-based design is limited by scarce experimental complex structures. Pearl's synthetic-data training is aimed at that gap.

3. Competitive Landscape

Player Approach Vs. Genesis
isomorphic-labs AlphaFold-lineage models, pharma deals plus own pipeline Same structure-model-led strategy at larger scale
iambic-therapeutics AI small molecules with weekly in-house design-make-test loops; clinical-stage Further along clinically; heavier in-house automation
chai-discovery Structure/design models sold to pharma Pure model vendor; Genesis also owns assets
schrodinger Physics + ML software plus pipeline Incumbent physics stack that Genesis's models aim to replace

4. Unique Observations

  • Partner cash replaced venture rounds. Since the 2023 Series B there has been no priced venture round. Funding has come from pharma upfronts and strategic equity: undisclosed from NVIDIA, $40M from Incyte [1][2][7].
  • The rename tracks the business. Moving from "Therapeutics" to "Molecular AI", and hiring a former Meta Llama lead as CTO [5], signals that the model is now the main product. That is consistent with partnership revenue far exceeding pipeline progress after seven years.
  • Lab-in-the-loop by contract. Rather than scaling its own robotic lab, Genesis has written partner data-sharing into the Incyte deal [2], an alternative to the heavy in-house automation route.

5. Financials / Funding

  • Seed ~$4M (2019, a16z; single source) [8]; Series A $52M (2020, led by Rock Springs Capital) [7].
  • SAFE notes $23.9M (2022), converted into the Series B [7].
  • Series B $200M new money (Aug 2023), co-led by a16z; NVentures, BlackRock, Fidelity and T. Rowe Price joined [6][7].
  • NVIDIA strategic investment (Nov 2024, undisclosed) and $40M Incyte equity (May 2026) [2].
  • Cumulative ≈ $300–340M. Third-party figure ~$296M before the Incyte equity; the company says "over $300M" [8]. Valuation is not disclosed.

6. People & Relationships

  • Co-founder & CEO: Evan Feinberg (Stanford PhD, Pande lab).
  • CTO: Sergey Edunov (joined Dec 2025; previously led Llama 2/3 research at Meta) [5]. Co-founder and former CTO Ben Sklaroff left in 2024 [8].
  • CSO: Shifeng Pan (ex-Novartis GNF San Diego discovery chemistry; ex-Odyssey Therapeutics).
  • Investors: a16z, Rock Springs, Menlo, Radical, nvidia (NVentures), Incyte. Partners: Incyte, Gilead.
