Kriya Therapeutics

Vertically integrated AAV gene-therapy company for common chronic diseases. It owns its own 3,000L GMP plant and has raised roughly $0.9B privately.

1. Core Product / Service

Kriya was founded in 2019 and launched in 2020 with dual headquarters in Palo Alto and North Carolina's Research Triangle; it is now headquartered in Morrisville, NC [1][2]. It develops AAV gene therapies, and some of its programs use "gene-encoded biologics" in place of chronic injections. Its targets are large indications rather than rare diseases.

The company says it has five programs in the clinic. All are Phase 1/2 per ClinicalTrials.gov [2]:

Program Indication Trial start
KRIYA-825 Geographic atrophy May 2025 (most advanced)
KRIYA-586 Thyroid eye disease May 2026
KRIYA-497 MASH July 2026
KRIYA-748 (chemogenetics + oral varenicline, from Redpin) Trigeminal neuralgia August 2026
KRIYA-288 Type 1 diabetes October 2026

Acquisitions: Warden Bio in January 2022 and Redpin Therapeutics in November 2022 [2].

2. Target Users & Pain Points

  • Patients: people with chronic diseases that today need frequent injections, such as GA and TED. A single gene-therapy dose could replace repeat dosing.
  • Pain addressed: manufacturing cost and scale have kept AAV confined to rare diseases. Kriya's argument is that owning large-scale manufacturing makes common indications economically viable.

3. Competitive Landscape

Player Approach Note
Apellis / Astellas Approved complement drugs for GA Chronic intravitreal injections that Kriya aims to replace
4D Molecular Therapeutics AAV for ophthalmology Engineered capsids, mostly retinal
Regenxbio / AbbVie AAV ocular (ABBV-RGX-314) Later stage in wet AMD

4. Unique Observations

  • Wet lab and manufacturing are its strategy, not just a cost line.
    • 2020: secured a 51,350 sq ft manufacturing facility in RTP [6].
    • July 2021: finished renovations, adding "multiple cGMP suites" up to 3,000L and "vial-to-vial" production [6].
    • Today: runs 50L, 500L and 3,000L GMP trains, in-house upstream and downstream process development, analytics and QC, and automated fill-finish. It reports more than 100 engineering and GMP batches at 50L or larger since 2019 [5].
    • June 2026: chosen as one of seven participants in the FDA's PreCheck pilot for a new commercial-scale facility in RTP [7].
    • Not disclosed: the CDMO or CRO names, and the square footage of the new plant.
  • Investors are paying for manufacturing. The Series D co-lead pointed to manufacturing constraints as what has held gene therapy back [3]. Among 2018–2022 non-AI biotechs, Kriya is the clearest case of a company that builds GMP capacity instead of renting it.
  • Headline totals overstate the cash. SEC Form D filings sum to about $870M sold. Aggregators show $927.6M because they count the full announced $150M 2023 extension, and some outlets cite "over $1.2B" [1][4].

5. Financials / Funding

All amounts below are from SEC Form D filings except where noted [1].

Round Date Amount Lead / notes
Series A 2020 $87.6M (press: $80.5M)
Series B June 2021 $100.0M
Series C May 2022 $270.5M Patient Square
Series C extension July 2023 Announced as $150M; Form D shows ~$91M drawn by then [4]
Series D August / September 2025 $313.3M at first filing, amended to $320.8M Patient Square and Premji Invest; also Peter Thiel, Narya, T1D Fund [1][3]
  • Cumulative: about $870M sold per SEC filings, or about $930M on the headline basis.
  • Status: private, with no IPO filing as of October 2026.

6. People & Relationships

  • Co-founder and CEO: Shankar Ramaswamy (ex-CBO, Axovant Gene Therapies) [2].
  • Vice chair: Roger Jeffs (ex-CEO, United Therapeutics) [1].
  • CMO: Greg Di Russo (since December 2025). CFO: Sachiyo Minegishi (since January 2026) [2].
  • Investors: Patient Square Capital, Premji Invest, Foresite, and others [1][3].
